Best adaptation in that the movie version was orders of magnitude better than the book: Jurassic Park.

Lots of good ones already mentioned, but I'd add Noises Off (yes, I know it's based on a play), Hitchcock's The Trouble with Harry and, though it may not qualify as pure comedy, the cult movie Harold and Maude despite the Cat Stevens soundtrack.

Just got back from seeing it -- excellent film! Gritty, dark, well acted. I have a couple of gripes, most notably the stupid apparently obligatory car chase scene, but overall I was very, very pleased. Now that they have Batman down they should move on and do Grendel.
